Specificity Statement:This antibody is capable of recognizing histone H3 proteins with a trimethylation at Lys27. It specifically recognizes and binds trimethyl lysine and amino acids surrounding it comprising KQLATKAAR(trimethyl K)SAPATGGVKC. Methylation is a type of post-translational modification (PTM) which is caused by covalent addition of methyl group to proteins, DNA or other molecules. PTMs have fundamental roles in chromatin structure and function. The PTMs that have taken place in histone proteins can affect gene expression by altering chromatin structure. Histone modifications have also been known as epigenetic markers. Defects in the PTMs pathway have been associated with the occurrence and progression of various human diseases, such as cancer, heart failure, autoimmune diseases, and neurodegenerative disorders such as Parkinson's disease, Alzheimer's disease, and Huntington's disease.
Application Notes:This antibody was used for the development of a H3K27me3‐specific intracellular antibody or H3K27me3‐mintbody, which was a fusion protein comprising variable domain regions of 2E12LI antibody fused to a fluorescent protein and expressed in mouse teratocarcinoma cell line MC12. This mintbody was capable of tracking H3K27me3 in living cells. Mouse MC12 cells, which stably express H3K27me3‐mintbody (sfGFP) was used to localize H3K27me3‐enriched chromatin in living cells. The binding specificity of bacterially expressed and purified 2E12LI‐sfGFP in vitro was evaluated using a modified histone peptide array. (PMID: 33605056).
