| Chimeric Use Statement: | This chimeric rabbit antibody was made using the variable domain sequences of the original Human Fab format, for improved compatibility with existing reagents, assays and techniques. |
| Specificity Statement: | This antibody is specific for human and cynomolgus monkey CNN1. |
| Application Notes: | The original antibody was used for flow cytometry on suspension adapted HEK293 cells. In a further flow cytometry experiment it was shown that the antibody also bound to cynomolgus monkey CNN1 This antibody was used for an ELISA on both human and mouse CNN1. The binding affinity of this antibody to human CNN1 was determined using surface plasmon resonance. It showed a Kd of 3.02 * 10^-8. This antibody was used for immunofluorescence on NIH 3T3 cells or suspension adapted HEK293 cells transfected with the expression plasmids driving the expression of the CCN1. It showed it could correctly label the CNN1 protein in vitro. This antibody was used in vivo on different tumor cell lines xenografted into a SCID-beige mice. It was effective for SKOV3 cells and Panc-1 cells but not MDA-MB-435 (US20120258112A1). |
