Specificity Statement:The antibody targets the central region of amyloid beta (residues 17–22).
Application Notes:The specificity of the original, Fab and scFv formats of the antibody was confirmed by ELISA analysis (Tammer et al., 2002; PMID: 12197886, Nisbet et al., 2013; PMID: 23766374). The original format of the antibody is extensively used for immunohistochemistry for detection of Aβ plaques. The original format and the Fab fragment bound to Ab[1–40/42/43] peptides by western blot analysis. The Fab format of the antibody shown to bind plaques within sections of human AD brain tissue by immunohistochemistry (Tammer et al., 2002; PMID: 12197886). The scFv format of the antibody bound to the central region of Amyloid beta with an affinity of ∼55 nM and significantly reduced fibril formation of Amyloid beta 1–42. Furthermore, the scFv format of the antibody ameliorates Ab1–42-mediated toxicity in the PC12 cell line and murine primary neuronal cultures (Nisbet et al., 2013; PMID: 23766374). The antibody was able to immunoprecipitate aliquots of individual EDTA-blood plasma samples from five different donors by magnetic bead immunoprecipitation (Klafki et al., 2020; PMID: 32911706).
