The original antibody was generated by immunizing rabbit with a synthetic peptide comprising amino acid sequence 'SIRLQI(pS)NPDLKDC'. The immunogen sequence was phosphorylated at the 7th serine residue which corresponds to serine 446 of human IRF5. A phage display scFv library was generated using the cells from the spleen and the specific antibody was isolated by positive screening against biotinylated 'SIRLQI(pS)NPDLKDC' peptide and negative selection biotinylated non-phosphorylated peptide 'SIRLQISNPDLKDK'.

Chimeric Use Statement:This full-length, chimeric human antibody was made using the variable domain sequences of the original Rabbit scFv format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds human and mouse IRF5 phosphorylated at serine 446. IRF5 is a transcription factor that plays a critical role in innate immunity by activating expression of type I interferon (IFN) IFNA and INFB and inflammatory cytokines downstream of endolysosomal toll-like receptors TLR7, TLR8 and TLR9.
Application Notes:The binding evaluation of this antibody to human and mouse pS446-type IRF5 was done using ELISA. This antibody was used in the detection of endogenous IRF5 upon TLR stimulation of human immune cell lines. Phosphorylation of endogenous IRF5 in CAL-1 cells stimulated with R-848, a ligand for TLR7 and TLR8, was analyzed by immunoblotting. Similar method was used for the detection of endogenous IRF5 phosphorylation upon TLR stimulation of human PBMCs. This antibody was also used for the determination of phosphorylated IRF5 positive cell population in human PBMC using flow cytometry. The antibody was also able to detect phosphorylated mouse IRF5 protein. This antibody was also used in the phosphorylated IRF5 from splenic CD11c cells of 18-20 week old female Lyn-deficient mice and NZB/W F1 mice, a widely used SLE model mouse.
