Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody is specific for human CD39, also known as NTPDase 1 (ectonucleoside triphosphate diphosphohydrolase) or vascular CD39. CD39 works in concert with CD73 (ecto-5'-nucleotidase) to hydrolyze extracellular adenosine triphosphate (ATP) and adenosine diphosphate (ADP), generating adenosine. The resulting adenosine binds to adenosine receptors, inhibiting T-cell and natural killer (NK)-cell responses, thus suppressing immune system activity. Additionally, CD39 may play a role in preventing platelet aggregation by converting platelet-activating ADP into AMP.
Application Notes:The specificity of the original version of this antibody (Human IgG1, Fc Silent) was assessed using ELISA. Among the tested targets — vascular CD39 and the CD39 isoforms L1, L2, L3, and L4 — the antibody exclusively bound to vascular CD39. In a flow cytometry (FC) assay using Ramos human lymphoma cells, which express CD39, the antibody demonstrated strong binding. Additionally, the antibody exhibited potent inhibition of CD39 activity, as measured by a MALDI-TOF (Mass Spectrometry) assay, effectively blocking over 90% of CD39's enzymatic activity (US20220056145A1).
