| Application Notes: | The original antibody (Mouse IgG1) demonstrated high-affinity binding to CHO cells expressing human CD96 isoform 2 (EC50 = 5.9 ng/mL), as validated in an ELISA. It effectively blocked CD155-Fc binding to the same target (IC50 = 152.4 ng/mL) in a competitive binding assay. It also exhibited binding to CHO cells expressing the V/V variant of human FcγRIIIa (EC50 = 191.5 ng/mL), confirmed through an ELISA. In an in vitro T cell activation assay, the antibody successfully bound to activated CD4+ and CD8+ T cells from three donors, and in a staphylococcal enterotoxin A (SEA)-stimulation assay, it showed limited functional activity in promoting IL-2 secretion by PBMCs. Additionally, it displayed a binding affinity (KD = 13.3 nM) for the full-length extracellular domain of human CD96 in a kinetic analysis (US20240141039A1). |
