Chimeric Use Statement:This chimeric rat antibody was made using the variable domain sequences of the original Human IgM format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:The original antibody binds to the disialylated gangliosides GD1b, GD3, GT1b, and GQ1b and weakly to GM3 and GD1a. It does not bind to structurally dissimilar glycolipids, including GM1, GM2, globoside, and sulphatide.
Application Notes:The original format of the antibody bound to a wide array of structures in the rodent peripheral nervous system, including dorsal root ganglia, peripheral nerve axonal profiles, compact myelin, nodes of Ranvier, motor end plates, and muscle spindles. In both rat and mouse dorsal root ganglia, the original format of the antibody stained the plasma membrane and cytoplasm of neurons. The antibody staining extended into the axonal lumen of myelinated nerve fibers. In teased fibers from mouse sciatic nerve, antibody-labeled compact myelin exposed at points of damage, created during the teasing process, and labeled axons protruding beyond ruptured myelin sheaths. It also labeled nodes of Ranvier in some intact teased fibers. In both mouse and rat, the original format of the antibody strongly labeled capillaries within the nerve bundle. In the rat muscle spindle, the antibody bound strongly to a region immediately adjacent to the intrafusal fibers, corresponding to the area defined by the internal capsule. The original format of the antibody also bound in the region of the neuromuscular junction in both mouse and rat muscle, as demonstrated by colocalization of Ha1 with BTx, a specific ligand for postsynaptic acetylcholine receptors (Willison et al., 1996; PMID: 8636426).
