| Application Notes: | The specificity of the original format of the antibody was confirmed by yeast surface display assay (Kd= 6,5 nM). The antibody, together with 4-5 was employed to build an heterodimeric clasping antibody with enhanced binding properties. The clasping antibody showed Kd= 1.2 nM by peptide IP assay. Heterodimeric clasping antibodies in an IgG like format bound to H3K27me3 in a IP assay (Kd= 2.1 nM). Phosphorylation of the H3K27me3 peptide at S28 moderately reduced the affinity of clasping antibody (Kd=40 nM). Biolayer interferometry measurements revealed a very slow dissociation rate of clasping antibodies (Kd=2.1 nM). The clasping antibody detected e histone H3 protein in the whole-cell lysate (WCL) by western blot analysis (Riso et al., 2025 ; PMID: 39793060). |
