Application Notes:The specificity of the original format of the antibody was confirmed by yeast surface display assay. The original format of the antibody specifically captured nucleosomes harboring H3K27me3 by Internal Standard Calibrated chromatin immunoprecipitation. The antibody, together with 4-5 was employed to build an heterodimeric clasping antibody with enhanced binding properties. The scFv format of the antibody, together with 4-5 was selective for H3K27me3 in an IP assay. The clasping antibody showed Kd= 0.4 nM by peptide IP assay (Kd= 1.8 nM). Heterodimeric clasping antibodies in an IgG like format bound to H3K27me3 in a IP assay (Kd= 2.1 nM). Phosphorylation of the H3K27me3 peptide at S28 moderately reduced the affinity of clasping antibody (Kd=7.4 nM). Biolayer interferometry measurements revealed a very slow dissociation rate of clasping antibodies (Kd= 1.1 nM). The clasping antibody detected e histone H3 protein in the whole-cell lysate (WCL) by western blot analysis. Quantitative native ChIP incorporated with synthetic nucleosomes revealed the long-neck scFv-Fc clasping antibody specifically captured nucleosomes harboring H3K27me3 (Riso et al., 2025 ; PMID: 39793060).
