Specificity Statement:This antibody binds 2,4 dinitrophenol (DNP) and also cross reacts with various other DNP haptens like DNP-TEMPO-ethylenediamine (DNP-SL), DNP-TEMPO-glycine (DNP-SG), DNP-bis(glycine) (DNP-G2), DNP-TEMPO-aspartic acid (DNP-SA), DNP-bis(aspartic acid) (DNP-D2), DNP-bis(ethylenediamine) (DNP-en2), DNP-glycine (DNP-G). Anti-DNP antibodies can be used as an independent quality control system in immunochromatography which can improve the specificity of C-line. They also have applications in visualization of DNP-labeled nucleic acid probes in techniques like in situ hybridization (ISH) and immunohistochemistry (IHC). They also serve as isotype controls in flow cytometry, immunofluorescence, and other assays to validate specificity.
Application Notes:The binding of this antibody to DNP and other related haptes was checked using ELISA. AN02 and related antibodies were engineered for nuclear magnetic resonance (NMR) to enable sequence-specific resonance assignments, aiding in structural analysis of antibody-antigen interactions (PMID: 3375235). The crystal structure of AN02 Fab fragment complexed with DNP-hapten was resolved at 2.9 Å resolution using X-ray crystallography. It was reported that the DNP ring was sandwiched between tryptophan residues Trp96 of the heavy chain and Trp91 of the light chain (PMID: 1920408).
