| Specificity Statement: | This antibody binds 2,4 dinitrophenol (DNP), also binds related DNP analogs like 2-nitrophenol (NP) and 2-nitro-4-iodophenol (PMID: 12610298) and some unrelated compounds like alizarin red (Az, Kd=40 nM) and furazolidone (Fur, Kd= 1.2M) (PMID: 1650428). Anti-DNP antibodies can be used as an independent quality control system in immunochromatography which can improve the specificity of C-line. They also have applications in visualization of DNP-labeled nucleic acid probes in techniques like in situ hybridization (ISH) and immunohistochemistry (IHC). They also serve as isotype controls in flow cytometry, immunofluorescence, and other assays to validate specificity. |
| Application Notes: | This antibody is one of the most widely used mouse IgE to study IgE biology and function. The original mouse IgE antibody binds DNP with an affinity of 20nM. Its affinity for close analogs of DNP, such as 2-nitrophenol (NP) and 2-nitro-4-iodophenol, is negligible (Kd >100 µM). The binding characterization of this antibody was done using ELISA. SPE-7 is reported to have the ability to trigger mast cell activation and survival independent of a cross-linking antigen (PMID: 28811536; 12360215) the potential to bind DNP-unrelated antigens through conformational diversity (PMID: 12610298) and an increased capability to bind mouse Fcγ receptors (PMID: 17558411). |
