Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds human and cynomolgus monkey tryptase, which is a major neutral protease present in mast cells and is secreted upon the coupled activation-degranulation response of this cell type. Elevated tryptase levels are found in subgroups of patients with mastocytosis, myelodysplastic syndrome, myeloproliferative neoplasm, acute myeloid leukemia, chronic myeloid leukemia (CML) and chronic eosinophilic leukemia (CEL). It could therefore serve to have diagnostic or prognostic significance.
Application Notes:The binding affinity of this antibody for human and cynomolgus monkey tryptase was done using SPR. The original human IgG of this antibody binds human hTPSB1 and monkey mfTPSB1 with a binding affinity of 3.3X10^(-10) M and 4.3X^10(-8) M respectively. The original antibody was capable of depolymerizing human and monkey tryptase tetramers into monomers and combine with monomeric proteins to form antibody-antigen complexes. This antibody can effectively inhibit the activity of human and monkey trypsin-like enzymes in a trypsin-like enzyme activity assay based on the S-2288 substrate chromogenic reaction, with IC 50 values of 1.43nM and 0.48nM respectively. This antibody was also capable of inhibiting the proliferation of Ba/F3-PAR2 cells stimulated by β-type human trypsin (CN118324919).
