The antibody was generated by immunizing Sprague Dawley rats and half-lop rabbits with various antigens, including synthetic peptides mapping to regions common to both TcdA and TcdB full-length toxins, formaldehyde-inactivated toxoid A, binding domain fragments of Toxin A, binding domain fragments of Toxin B, or combinations of these. Following immunization, B cells were isolated, and the resulting antibodies were subsequently humanized.

Specificity Statement:This antibody is specific for Toxin A from Clostridium difficile.
Application Notes:This antibody was used for in vitro neutralization assay on CACO-2 cells with toxin A, it was shown that this antibody has a high potency as neutralization agent, it had a substantial potential to neutralize TEER loss with maximal inhibition approaching 80% and an EC50 of approximately 80 ng/ml. This antibody was used for surface plasmon resonance on the subdomains of TcdA123 and Tcd456, it showed it was able to bind at least 12 times to Toxin A with an affinity of 132 pM. This antibody was used for an in vivo hamster infection study, it was able to improve survivability (Davies et al., 2013; PMID:23324518).
