| Application Notes: | The original antibody was used to block PtdSer binding to TIM3. Jurkat E6-1 cells were treated with paclitaxel to induce apoptosis and promote the accumulation of PtdSer on the cell surface. Then, it was mixed with mFc-labelled TIM3, this antibody, and control antibody. The TIM3 protein bound to PtdSer on the surface of apoptotic cells was detected with PE-labelled anti-mouse Fc antibody using flow cytometry. It showed that this antibody could inhibit binding of TIM3 to PtdSer in a dose-dependent manner.The affinity of this antibody to TIM3 was determined using surface plasmon resonance, it showed a affinity of 3.64*10^-5 Kd . The functional activity of this antibody was determined by measuring IFN-γ secretion by DC-T cell co-culture assay, it showed that it could increase secretion of T-cell IFN-y (CN112062850B). |
