Chimeric Use Statement:This is a reformatted mouse IgG2b Fc Silent™ antibody, based on the original mouse scFv format, created for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds human and mouse FLT3 (CD135). FLT3 is a tyrosine-protein kinase that acts as a cell-surface receptor for the cytokine FLT3LG and regulates differentiation, proliferation and survival of hematopoietic progenitor cells and of dendritic cells. FLT3 mutations, particularly internal tandem duplications (ITD) and tyrosine kinase domain (TKD) mutations, are frequently found in acute myeloid leukemia (AML) and are associated with a poorer prognosis. These mutations can drive the development and progression of AML by promoting the uncontrolled proliferation of leukemic cells.
Application Notes:This antibody binds the extracellular domain of the recombinant human and mouse FLT3 in an ELISA. It inhibits binding of FLT3 ligand to recombinant human and mouse full-length FLT3 expressed in HEK293 cells. This antibody antagonizes FLT3 ligand-induced FLT3 activation in RS4-11 cells. It inhibits FLT3 ligand-induced FLT3 autophosphorylation at concentrations of 10 and 100 μg/ml. It was reported to be potent at antagonizing native FLT3 in sensory neurons and efficacious in inhibiting neuropathic pain in a Chronic Constriction Injury (CCI) murine model (US10781452).
