| Application Notes: | The specificity of the original format of the antibody was confirmed by competitive ELISA. The original format of the antibody was used in a competition experiments on rabies virus G protein (ERA strain) transfected PER.C6 cells by means of flow cytometry. The signal obtained with the biotinylated antibody was not reduced significantly with the negative control antibody. Competition with the unlabeled antibody reduced the signal to background levels. The binding affinity of the scFv fragments to purified rabies glycoprotein was measured by surface plasmon resonance (Kd= 2.4 nM). The neutralizing potency of the original format of the antibody to rabies G protein IgGs was determined using an in vitro neutralization assay (Modified RFFIT). The antibody could neutralize anti-rabies virus G protein with a potencies of 3800 IU/mg (US7579446B2). |
