| Application Notes: | The specificity of the original format of the antibody was confirmed by ELISA analysis. The original format of the antibody inhibited the binding of human VEGF to human VEGF receptor Flt-1 with a ratio of 66.3%. The original format of the antibody inhibited binding of human VEGF to human VEGF receptor Flt-1 in a concentration dependent manner (IC50 = 2.0 μg/ml). The original format of the antibody inhibited binding of human VEGF to NIH3T3-Flt-1 cells in a concentration dependent manner (IC50 = 0.041 μg/ml). The original format of the antibody specifically recognize the human VEGF receptor Flt-1 on the NIH3T3-Flt-1 cell surface by flow cytometry. The original format of the antibody could detect human VEGF receptor Flt-1 on human vascular endothelial cells. The effect of the original format of the antibody on the VEGF-dependent migration activity of human vascular endothelial cells, as an index of in vitro angiogenesis activity, was examined. The study revealed that cell migration ability of HUVEC increased by adding VEGF, but the migration was completely inhibited by adding the antibody. Further, the antibody showed the activity to inhibit the migration of vascular endothelial cell concentration-dependently in the range between 0.1 and 1 μg/ml (US7615214B2). |
