Quick Overview
The original antibody was generated by immunizing XenoMouse® with human uPAR expressed on the surface of B cells. Recovered lymphocytes from hyperimmunized mice are fused with a myeloid-type cell line to prepare immortal hybridoma cell lines. Antibodies from the cell supernatants are then screened by ELISA for reactivity against the uPAR immunogen.

Chimeric Use Statement:This is a reformatted human IgG1 Fc Silent Fc Silent™ antibody, based on the original human IgG2 format, created for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds human urokinase plasminogen activator receptor (uPAR). It also cross reacts with cynomolgus monkey UPAR. It plays a role in localizing and promoting plasmin formation. Mediates the proteolysis-independent signal transduction activation effects of U-PA. Upregulation of uPAR expression at the cell surface, and plasminogen activation, has been linked to a number of conditions including inflammation, wound repair, arthritis, atherosclerosis angiogenesis, tumor invasion and metastasis.
Application Notes:This antibody inhibits uPA binding to the cell surface of U937 cells. In an adhesion assay, this antibody inhibited cell adhesion of U937 cells to vitronectin in a dose-dependent manner. At a concentration of 2 μg/ml, 2.6.1 achieved 90% of inhibition. The binding characterization of this antibody to human UPAR was done using High Resolution Biacore™. The binding affinity of the original IgG isotype for human UPAR was reported to be Kd= 249pM. The binding characterization of biotin conjugated version of this antibody was done using immunohistochemistry on frozen cell pellets and normal human tissue microarrays (US8026344).
