Chimeric Use Statement:This full-length, reformatted human antibody was made using the variable domain sequences of the original Human scfv format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds granzyme B, an abundant serine protease in the cytosolic granules of cytotoxic T-cells and NK-cells which activates caspase-independent pyroptosis when delivered into the target cell through the immunological synapse. It has has been demonstrated to be a pivotal molecule for protective immunity against viral infection and cellular malignant transformation.
Application Notes:This antibody can detect the native for of granzyme B and fails to detect its denatured form. This antibody can be used as a tool to monitor granzyme B expression in natural killer cells and antigen specific CD8 T cell immunity by intracellular staining using FITC-labelled Fv17 together with PE-labelled anti-perforin IgG. Stained cells were analyzed by flow cytometry. This antibody failed to detect granzyme B immobilized on plastic in a direct ELISA. This antibody in combination with other commercially available anti-granzyme B antibody BG10 can be used for sandwich ELISA. This antibody can also be used to detect granzyme B and granzyme B-PI9 conjugate using immunoprecipitation (PMID: 15319069).
