| Application Notes: | The original format of the antibody bound human and murine Robo4 expressed endogenously in human HUVEC cells and murine MS1 cells by FACS analysis. The original antibody inhibited HUVEC tube elongation. Binding affinities of the antibody to human and mouse Robo4 were determined by SPR measurements for the IgG1 format of the antibody (KD= 1.1 nM and 0.9 nM for human and mouse respectively), the IgG1 format of the antibody with engineered Cys (KD= 1.2 nM and 1.1 nM for human and mouse respectively), and a Fab fragment with engineered Cys (KD= 8 nM and 1.5 nM for human and mouse respectively). Binding affinities of the antibody to human and mouse Robo4 were determined by radiolabeled cell binding for the IgG1 format of the antibody with engineered Cys(KD= 1.2 nM and 1 nM for human and mouse respectively), and a Fab fragment with engineered Cys(KD= 3.2 nM for mouse) (US7834154B2). |
