| Application Notes: | The original format of the antibody was able to inhibit the cell growth of HEK-OX1R. The original format of the antibody reduced the HEK-OX1R cell number of about 46%. The inhibition of cellular growth induced by the original format of the antibody was totally reversed by an inhibitor of SHP2 protein, demonstrating that the cellular growth inhibition mediated by the orignal format of the antibody was associated to the SHP2 signaling pathway. The original format of the antibody promoted apoptosis of CHO cells lines transfected with OX1R of mouse or rat. The original format of the antibody could inhibit the cellular growth in cancer cell lines derived from colon cancer (SW48, LoVo) and pancreas cancer (AsPC-1 cells). Further, the original format of the antibody induced apoptosis in HEK-OX1R cells. The ability of the original format of the antibody to interact with the OX1R binding site was determined by competitive inhibition of 125I-OxA binding study. The antibody was able to competitively inhibit the binding of 125I-OxA to HEK-OX1R with an estimated IC50 of about 5μM (US20180030135A1). |
