Application Notes:The specificity of the original format of the antibody was confirmed by competitive ELISA. hIGF-I and hIGF-II were inhibited by the original format of the antibody. The binding affinity of the original format of the antibody to hIGF-I and hIGF-II was measured by surface plasmon resonance (Ka = 7.86×10^9 M^−1 and 8.63×10^9 M^−1 for hIGF-I and hIGF-II respectively). The original format of the antibody strongly inhibited the human breast cancer cell line MCF7 growth by hIGF-I and hIGF-II. The original format of the antibody inhibited the growth of a human colon cancer cell strain HT-29 and the growth of a human osteosarcoma cell strain MG63 by hIGF-I and hIGF-II. The original format of the antibody could inhibit the autocrine cell growth by the hIGF-I produced by the cell itself. The original format of the antibody could inhibit the anchorage independent growth of hIGF-I expressing cells. The original format of the antibody inhibited the growth of tumors in vivo due to the inhibition of hIGF-I (US7438911B2).
